Media Compounding
Weigh, dissolve, adjust pH, and sterilise a batch of medium.
4 steps · Reagent preparation · Media prep
1. Weigh and dissolve the dry components
Weigh each component into roughly 80% of the final volume of water and stir until fully dissolved before topping up.
2. Adjust pH and bring to final volume
Adjust to the target pH, then top up to the final volume. Adjusting after topping up changes the concentration.
3. Autoclave
Sterilise on a liquid cycle. Loosen caps — a sealed bottle is a pressure vessel.
4. Cool, then add heat-labile supplements
Cool to below 55 °C before adding antibiotics or other heat-labile components, then record the lot on the bottle.
